I know a lot of people are already familiar with using JPG, but I thought I would add this suggestion that if you are going to be saving a photograph for posting, that you use a quality level of at least 80%. The higher the compression, the more artifacts get introduced, and they often detract from the image. I'm so used to seeing them that they jump right out at me and can ruin an otherwise great shot. In the days of 2400 baud modems and limited disk space, small image file sizes were really important. Now, with dial-ups modems being more than 20x as fast, cable often being 1000x faster, and huge hard drives, I don't see much need for highly compressed JPGs, especially at the dimensions used for posting to the forum.agreed. Can someone tell me the safety precautions needed to be taken while handling inseticides? Even though I use a mouth mask, the spray seems to bother me and gives me a headache. new 14.4v x2 drill makes gear grinding sounds after releasing trigger on either speed range, and at any clutch setting. this happens every time with no load. is this normal or do i have a defective drill out of box?Seems normal. When I drive on the highway my car swerves a little bit. It feels unstable and wobbly like the wheels are going to fall off or something. Hi, I've been reading around about filters. But I'm a little confusticated. (I use film, not digital) I sometimes notice a yellow cast to indoor photo's taken where incandescent light is present. I've found this about it. Standard light bulbs are much warmer than daylight, only about 2900 K for a 100-watt light bulb. In the film days, you would get a very pronounced yellow cast without a blue filter over the lens (Kodak says 80A + 82B). But why would I need two filters instead of one? Hi, im using a stragene QikChange II site directed mutagenesis kit to do a site-directed mutagenesis study. The plasmid im using is pGL3-basic. It is about 4.8kb + my insert 500bp approximately = 5.3kb. So i did a PCR reaction at 95oC at 30secs.55oC at one minute and 68oC for 6 minute ( to play safe?) for 17 cycle. No band was observed after the cycling when i run on gel. But i continued with my Dpn I digestion as per protocol. But i did not get any colonies. I tried using different concentration of plasmid 10ng-50ng ( nothing was observed.) I used 1-2ul for my transformation - no colonies. How do You repair a broken 3/4" iner core cable? I haven't broke one ,but good to know! I am a very experienced welder. Mild steel Allum. stainless ,ect.with a 3/4 cable splicer ;)You could use a 3/4 inch splicer or you could put new ends on it. I usually like to do ends. If it is kinked severly I will cut out the kinked section, the grind the end of the cut flat. The cables I use come from the company I work for and have a plastic inner-core, so I try to grab it with a pair of needle nose and pull about a half inch out to make room for my splice. I then use loctite cleaner and clean the cable a bit and add green loctite to the screw in part of my spartan ends. Hi, I've been reading around about filters. But I'm a little confusticated. (I use film, not digital) I sometimes notice a yellow cast to indoor photo's taken where incandescent light is present. I've found this about it. Standard light bulbs are much warmer than daylight, only about 2900 K for a 100-watt light bulb. In the film days, you would get a very pronounced yellow cast without a blue filter over the lens (Kodak says 80A + 82B). But why would I need two filters instead of one? Hi, im using a stragene QikChange II site directed mutagenesis kit to do a site-directed mutagenesis study. The plasmid im using is pGL3-basic. It is about 4.8kb + my insert 500bp approximately = 5.3kb. So i did a PCR reaction at 95oC at 30secs.55oC at one minute and 68oC for 6 minute ( to play safe?) for 17 cycle. No band was observed after the cycling when i run on gel. But i continued with my Dpn I digestion as per protocol. But i did not get any colonies. I tried using different concentration of plasmid 10ng-50ng ( nothing was observed.) I used 1-2ul for my transformation - no colonies. How do You repair a broken 3/4" iner core cable? I haven't broke one ,but good to know! I am a very experienced welder. Mild steel Allum. stainless ,ect.with a 3/4 cable splicer ;)You could use a 3/4 inch splicer or you could put new ends on it. I usually like to do ends. If it is kinked severly I will cut out the kinked section, the grind the end of the cut flat. The cables I use come from the company I work for and have a plastic inner-core, so I try to grab it with a pair of needle nose and pull about a half inch out to make room for my splice. I then use loctite cleaner and clean the cable a bit and add green loctite to the screw in part of my spartan ends.
